1. Brief

4. Principle
The
Aflatoxins B1 rapid test strip is based on competitive inhibition immuno-chromatographic principle. In the flow process, Aflatoxins B1 in the
sample combined with Aflatoxins B1 specific colloidal
gold-labeled monoclonal antibody, Inhibit the combination between antibody and AFB1
-BSA conjugate on Test line of NC membrane, lead to the color change of Test
line.
When
the sample has no Aflatoxins
B1 residue or concentration lower than
detection limit, T line is darker than or has same color with C line. when the
concentration is equal to or higher than detection limit, T line is lighter
than C line obviously or T line is invisible. No matter whether there is Aflatoxins
B1 residue in sample, C line will
appear, it means the test is valid.

6. Sample
preparation
6.1 A. Grain: rice, wheat, corn, sorghum
B. Feed: regular feed, DDGS, soybean meal
C. Beans: peanut, soybean
Weigh crushed sample 1±0.05 g into 15ml centrifuge tube, add 4ml B1
Sample extracting buffer, shake vigorously by hand or Vortex for 1-2 min, centrifuge at 4000r/min at room temperature
for 5min, Take up-layer clear liquid to test.
6.2 Special sample ( sample is highly
absorbent)
Wheat bran, rice bran, green fodder
Weigh crushed sample 1±0.05 g into 15ml centrifuge tube, add 10ml B1
Sample extracting buffer, shake vigorously by hand or Vortex for 1-2 min, centrifuge at 4000r/min at room
temperature for 10min, Take up-layer clear liquid to test.
6.3 Edible oil
Use Pipette to take 200ul sample into 50ml or
15ml centrifuge tube, add B1 Sample extracting buffer(Quantity as following
tablet),
shake vigorously by hand or Vortex for 3 min,take sample to test.

7. Operation
procedures
7.1 Read the instruction
carefully before use. Return test strips and sample into room temperature.
7.2 Take bottles needed
from the kit package, take out required micro-wells and strips, making proper
marks. Please use these test strips within 1h. Seal the cap of the bottles,
avoid moisture.
7.3 Take 200ul of the
test samples into the micro-wells, then repeatedly absorb for 5 times, mix the
sample with the reagent in the micro-wells completely until no solid judge by
eyes (this is a very important step), Incubate for 5min at room
temperature(20-25℃).
7.4 Put test card on
desk flatly, drop 80µl liquid from the micro-well, add into the “S”hole on test
card.
7.5 Wait for 5-8min then
read the result, it is invalid in other time.
8. Test
Result Interpretation
8.1 Negative: T line is
darker than or has same color with C line. It means there is no Aflatoxins B1
residue in sample or the residue is lower than detection limit.
8.2 Positive: T line is
obviouly lighter than T lin or T line is invisible. It means the Aflatoxins B1
residue is higher than or equal to detection limit.
9. Precautions
1) The test strips can
be used only once at room temperature, do not use test strips out of expiry
date.
2) Please seal the bottle after taking out
required test strips. If can not use all 8 micro-wells at once, cup the rest
and put it back to bottle and sealed.
3) Do not touch the
white membrane surface in the middle of test strips, avoid sunlight and fan
blowing directly.
4) Use the test strips
testing again for positive results.
5) Please contact the
supplier for any questions.
6) The result is for
reference only, for confirmation, please refer to the relevant national
standard method.
10. Specificity
Test 500ppb of
Tetracyclines, Chloramphenicol, Sulfonamides etc., the result is Negative.
11. Storage
and expiry date
Storage: Store at 2-8 ℃ in dark,
sealed, dry place, no frozen.
