Principle
This rapid test strip applies the bioorganic
chemical colorimetric method and is suitable for detecting the content of β -
hydroxybutyric acid (BHB) in cow milk. The principle is that BHB in breast milk
reacts with coenzyme I (NAD) under the catalysis of BHB dehydrogenase to
produce acetoacetic acid and reduced coenzyme I (NADH); The generated NADH
reacts with nitroblue tetrazole (NBT) under the catalysis of Huangdi enzyme to
produce blue black insoluble formazan, and a standard color chart is made based
on the color intensity of the generated chromogenic substance.

3. Components
1). One white packaging bottle (with desiccant
inside);
2). 25 test strips;
3). Color chart (pasted on the bottle body);
4). One copy of the user manual
4. Operation procedures
1). Collect fresh cow milk in a container, and
the milk in the container should not exceed 1/3 of the test strip (the main
milk sample should be soaked in the reaction block). Collect and test on the
same day.
2) If on-site testing is not possible
immediately, the sample should be frozen and sealed at -80 ℃ for storage, and testing should be completed
within three months. When testing, please restore the sample to room
temperature before testing.
3). Take out the test strips from the 4 ℃ refrigerator, return to room temperature, take
out the required test strips from the bottle according to the number of test
samples, and quickly seal the bottle cap.
4) After immersing the test strip in the milk
sample for 30 seconds, wipe off any excess milk and compare it with the color
chart on the bottle body (please complete the comparison within 1 minute).

5. Result determination
The color depth of the test area at the reaction
end of the test strip changes with the variation of BHB content in breast milk.
After accurately reflecting for 30 seconds, compare the color of the test strip
testing area with the color chart on the bottle body. The judgment criteria are
as follows:
● Warning:
The color of the test area is darker than the
color block of 100 µmol/L (BHB) on the color chart, indicating a risk of
ketosis;
● Subclinical ketosis:
The color of the test area is darker than the
color block of 200 µmol/L (BHB) on the color chart;
● Clinical ketosis:
The color of the test area is darker than the
color block of 500 µmol/L (BHB) on the color chart.
6. Notice
1). A
single test strip is a disposable product. Do not reuse or use expired test
strips, do not fold or scratch the test strips, and do not touch the reaction
end of the test strips.
2). The
test strips and samples to be tested should be restored to room temperature
before use. After the test strips are unsealed, please use them as soon as
possible.
3). Water
or other liquids cannot be used as negative controls for testing.
4). If
expired, damaged, or contaminated test strips are found during purchase, please
replace them at the place of purchase.
5). During
the sample processing, please be cautious and wear protective gloves.
6). This
product is only for veterinary in vitro rapid diagnosis.
7). If
you encounter any problems during testing, please contact the supplier.
8). The
test results of this product are for reference only. If confirmation is
required, please refer to the relevant national standard methods.
9). When
color spots appear in the test area of the reaction end of the test strip, it
indicates that the test strip has failed; Please contact the supplier for
replacement.
7. Storage
and expiry date
Sealed, dry, and dark storage at 2-8 ℃; Do not freeze. The validity period is 18
months.
If stored at room temperature (25 ℃), sealed, dry, and away from light, the shelf
life is 4 months.
