[Brief]
This product is used for rapid quantitative
detection of Ochratoxin A (OTA) in grains and their products, feed raw materials, and
some finished feed products. It is suitable for on-site rapid detection by
various enterprises, grain depots, testing institutions, and government
regulatory departments.
[Principle]
This product is based on the principle of
competition and combined with fluorescence chromatography technology. The Ochratoxin
A (OTA) in the sample competes with the OTA-Ag coated on the NC membrane
testing line for fluorescent microsphere labeled OTA monoclonal antibody. As
the OTA content in the test sample increases, the amount of fluorescent
microsphere antibody complex bound to the testing line decreases, and the
fluorescence intensity of the testing line weakens accordingly. After the
reaction is completed, the content of OTA in the sample can be detected using a
matching handheld fluorescence immunoassay analyzer.

[Required equipment, consumables, and reagents] (Not provided)
1. Equipment and consumables: electronic
balance, oscillator, centrifuge, pipette, and tips (20-200μL,100-1000μL,
1-5mL), centrifuge tube (50mL, 1.5mL), disposable gloves and mask.
2. Reagent: Anhydrous ethanol (AR).
[Reagent preparation]
Sample extraction solution: Take 500mL of
anhydrous ethanol and add 500mL of pure water, mix well for usage.
[Experimental Steps]
1. Preparation before testing
a. Return the test card and sample diluent to room temperature (25℃) for at least 30 minutes.
b. Turn on the side power switch of the handheld fluorescence immunoassay
analyzer, enter the password 123456 and click the arrow in the bottom right
corner of the keyboard to complete the input. Click login to enter the main
interface of the instrument and preheat for 10 minutes.
c. Turn on the constant temperature timer, set the temperature to 37℃, and incubate for 8 minutes.
2. Sample preparation
Multiple point sampling is required for the test
sample, with a total sampling amount of 300-500g. After thorough mixing, the
particles can be homogenized to over 90% and pass through a 20 mesh sieve.
Extract the screened samples for testing.
a. Conventional samples
Weigh 5.0±0.05g of homogeneous sieved sample
into a 50mL centrifuge tube, add 25mL of sample extraction solution, shake for
3 minutes, and centrifuge at 4000rpm for 5 minutes. Take 100μL supernatant, add
700μL sample diluent, shake and
mix well for 10 seconds, ready for testing.
b. Bran sample
Weigh 2.5±0.025g of homogeneous sieved
sample into a 50mL centrifuge tube, add 25mL of sample extraction solution,
shake for 3 minutes, and centrifuge at 4000rpm for 5 minutes. Take 100μL
supernatant, add 700μL sample diluent,
shake and mix well for 10 seconds, ready for testing. (In this case,
multiplying the test result by 2 is the final concentration)
Note: When the concentration of the analyte in the
sample exceeds the range of the curve, the sample extraction solution can
be used to dilute the extracted supernatant by 2-5 times, then take 100μL
diluted extraction solution, add 700μL sample
diluent, mix thoroughly and add the sample for testing. The final result
is obtained by multiplying dilution factor.
3. Sample testing
a. Take out the warmed test card from the aluminum foil bag, place it on a
horizontal table, and use a pipette to aspirate 70μL of the diluted sample, add
into the sample hole, insert the test card into the constant temperature
timer socket, and incubate for 8 minutes;
b. Click on the "Test" module of the handheld fluorescence
immunoassay analyzer, insert the ID card into the ID card socket, click
"Select Project", click "Read Project Card", click
"Confirm", and the name of the read project will appear in the box
below "Select Project". After selecting the required testing project
and barcode, it will automatically return to the testing page.
c. After incubation, select the corresponding sample type curve according
to the instructions, insert the test card with the sample hole end facing
inward and left into the instrument, and click "Test" on the screen
for detection.
Note: During testing, do not point the
constant temperature timer socket towards the air conditioner or fan vent.

4. Result acquisition
The test results will be displayed on the screen
of the handheld fluorescence immunoassay analyzer. You can click to print to
obtain a paper printed report, or export an electronic version of the data
report through a data cable.
[Precautions]
1. Please use the test card within its validity
period, and each card can only be used once;
2. Please use the test card immediately when
opening the aluminum foil bag packaging;
3. Different batches of test cards, sample
diluents, and ID cards should not be mixed;
4. Do not touch the white film strip at the
detection window. If touched, please discard this card to avoid affecting the
detection results;
5. After incubating with the sample for 8
minutes, please test immediately and try to complete the test within 2 minutes;
6. During the testing process, reagents,
instruments, etc. should be avoided from direct sunlight, fans, and air
conditioning;
7. This product is only used for screening. If a
positive result is found, it is recommended to retest by HPLC or LC-MS;
8. If you encounter any problems during testing,
please contact the supplier.
9. The disposal of experimental waste shall be
carried out in accordance with national or local laws and regulations.
[Storage and shelf life]
1. This product should be stored in a dry and
dark place at 2-8 ℃, and should not be frozen.
2. The shelf life is 12 months. The batch number
and production date can be found in the outer packaging.
