I. Intended use

This product is used to detect African swine fever virus in the blood of suspected sick pigs and dead pigs.


2. Principle 

This product is based on colloidal gold rapid immunochromatography technique to detect ASFV in blood samples. After adding the sample to the “S” well on test card, if ASFV is present in the sample, the virus will specifically bind to the colloidal gold-labeled ASFV antibody to form a complex that moves along the chromatographic membrane and is captured by the pre-coated ASFV antibody on the chromatographic membrane. Then a wine red Test line is formed at the T-position of the cartridge. If there is no ASFV in the sample, no visible lines will be formed at the T line position. In addition, the system also designed a C line for verifying whether the test card is valid. The line should be colored regardless of the negative or positive result, otherwise it will be invalid.

5.Operation procedures

1) Read the manual carefully and return the test card and sample to room temperature(15~25℃) before use.

2)Detection of whole blood samples

2.1) Sample preparation: Use dropper to absorb fresh whole blood sample, drop 3-4 drops into Sample buffer tube. Use dropper to blow and beat repeatedly for 8~10 times to split all cells.

Notice 1: Whole blood samples should be freshly collected or EDTA anticoagulation samples which stored at 2~8℃for not more than 72 hours.

Notice 2: The Sample buffer tube contains sample buffer, and the liquid may stick to the cap or tube wall due to transportation bumps etc. Before opening the lid, force it down or centrifuge for 30 seconds to allow liquid to enter the bottom of the sample buffer tube.

Notice 3: Use dropper to absorb and blow out the liquid repeatedly to make the cells to be completely lysed, and the virus was released. This step directly affects the test results.

2.2) Reaction: Open the foil bag and take out the test card. Draw 3~4 drops of sample after the treatment in step 2.1) in “S” hole on test card quickly, in horizontal position and be still.

2.3) Read the results after 15~20mins, do not beyond 30mins.


6. Results


6.1Positive(+): Both on T Line and C line being seen red color reaction. It means there is ASFV antigen in sample;

Notice 4: The strength of the T lines will vary depending on the concentration of the ASFV antigen in the sample. Therefore, the T line of any red mark appears and can be judged as positive.

6.2Negative(-): no color reaction on Test Line (T Line),only on Control Line(C Line) being seen red color reaction. It means there is no ASFV antigen in sample or the levels is lower than the detection threshold;

6.3Invalid: No color reaction on C Line. It means the operation is wrong or the test card is invalid, test again.


7.Note

1) Do not use test card out of expiry date, use it in 1 hour after opening foil bag.

2) Avoid direct sunlight and direct blows during testing.

3) Try not to touch the white film surface of the non-shell plastic part of the test card.

4) Do not mix usage of dropper to avoid cross pollution.

5) Do not use Sample buffer from other sources.

6) The test card after usage should be treated as a microbiological dangerous product.

7) This product does not apply to serum or plasma samples.


8. Storage and expiry date

Storage:  Store at 2-30℃in dark, sealed, dry place, no frozen.

Expiry date: 12 months; date of production is on box.
